Therapeutic indication of the resolution

Keytruda is indicated in combination with pemetrexed and platinum-based chemotherapy for the first-line treatment of unresectable non-epithelioid malignant pleural mesothelioma in adults.

    • KEYNOTE 483 is a completed, open-label, randomised Phase II/IIItrial in which pembrolizumab in combination with pemetrexed and platinum-based chemotherapy (arm A) was compared with pemetrexed in combination with platinum-based chemotherapy (arm B) and with pembrolizumab monotherapy (arm C).

Adult patients with unresectable, non-epithelioid malignant pleural mesothelioma; first-line treatment

  • An additional benefit is not proven.
  • The data from the KEYNOTE 483 trial are not suitable for assessing additional benefit. The comparison arms of the trial consist of pemetrexed in combination with platinum-based chemotherapy and pembrolizumab as monotherapy. This does not correspond to the appropriate comparator therapy of nivolumab in combination with ipilimumab. Consequently, the appropriate comparator therapy has not been implemented, meaning that, on balance, there are no suitable data available for assessing the additional benefit of pembrolizumab in combination with pemetrexed and platinum-based chemotherapy.
  • It is therefore concluded that an additional benefit is not proven for pembrolizumab in combination with pemetrexed and platinum-based chemotherapy as first-line treatment for unresectable, non-epithelioid malignant pleural mesothelioma in adults.
